HoxA9 Antibody (HOX5I043) is a mouse monoclonal IgG1 kappa light chain antibody that detects HoxA9 protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). HoxA9 (HOX5I043) antibody is available as a non-conjugated anti-HoxA9 antibody. The HOX homeobox genes encode critical proteins involved in embryonic development, and HOXA9 specifically encodes a class I homeodomain protein expressed in both normal adult and fetal thymic tissue. This expression is vital as HOXA9 plays a significant role in regulating early differentiation of thymocytes, which are essential for developing a functional immune system. HOXA9 homeodomain protein cooperatively binds to consensus DNA sequences alongside Meis1 and Pbx1, forming a complex crucial for gene regulation during hematopoiesis. Notably, HOXA9 dysregulation, particularly with Meis1 and Pbx1, has been implicated in leukemic transformation in both mice and humans. HOXA9 and Meis1 overexpression in primary bone marrow cells induces growth factor-dependent acute myeloid leukemia (AML) in syngeneic mice, highlighting HOXA9′s importance in oncogenesis. Furthermore, chromosomal translocation t(7;11)(p15;p15) has been identified in patients with human AML and chronic myelogenous leukemia (CML), leading to NUP98-HOXA9 fusion gene formation. Mice transplanted with bone marrow cells expressing NUP98-HOXA9 develop a myeloproliferative disease (MPD) that progresses to AML, underscoring HOXA9′s critical role in both normal development and disease states.

I would like to know the desired dilution of this HoxA9 antibody (sc-81291) used in Western Blot-based experiments to detect murine Hoxa9. Also, at what molecular weight can we expect to see the band for murine Hoxa9?

Thank you for your question. As indicated on the product datasheet, HoxA9 (HOX5I043) is recommended for detection of HoxA9 of mouse, rat and human origin by Western Blotting (starting dilution 1:200, dilution range 1:100-1:1000). The expected molecular weight of HoxA9 is 36 kDa.

Would it be possible to get a small FREE 10 µg sample of HoxA9 (HOX5I043): sc-81291 so I can test in WB if it is worth to buy it. We tried sc17155 and did not work.

I am using HoxA9 (HOX5I043): sc-81291 antibody for immunoprecipitation, how much antibody should I use?

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
