Homer-1b/c Antibody (D-8) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 181-354 of Homer-1b/c of mouse origin, and specifically detects Homer-1b and Homer-1c proteins of mouse, rat, and human origin in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Homer proteins, also known as Vesl (VASP/Ena-related gene upregulated during seizure) or LTP, are a family of scaffolding proteins that play a crucial role in synaptic signaling by binding to group 1 metabotropic glutamate receptors (mGluRs), which are involved in intracellular calcium release. Structurally, Homer proteins contain an N-terminal Enabled/VASP Homology 1 (EVH1) domain that binds proline-rich sequences in target proteins and a C-terminal coiled-coil (CC) domain that mediates self-multimerization. This structure is essential for forming complexes and organizing signaling pathways at the postsynaptic density. Homer proteins facilitate synaptic plasticity and neurotransmission, which are critical for learning and memory. By linking mGluRs with intracellular calcium channels like inositol triphosphate receptors (IP3R), Homer proteins regulate calcium dynamics within neurons. Unlike the immediate early gene product Homer-1a, which lacks the coiled-coil domain and acts as a dominant-negative regulator, Homer-1b and Homer-1c are constitutively expressed and contribute to the stable formation of synaptic complexes.
