Homer Antibody (D-3) is a mouse monoclonal IgG1 kappa light chain antibody that specifically recognizes Homer protein of mouse, rat, and human origin in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Homer proteins, also known as Vesl proteins (VASP/Ena-related gene upregulated during seizure), are a family of scaffold proteins that play a crucial role in synaptic signaling by binding to group 1 metabotropic glutamate receptors (mGluRs), which are involved in triggering intracellular calcium release. The interaction between Homer proteins and mGluRs is essential for synaptic plasticity and neurotransmission, affecting learning and memory processes. Homer-1a, an immediate early gene product induced by neuronal activity, differs from other constitutively expressed Homer proteins (such as Homer-1b, -1c, -2, and -3) by lacking a coiled-coil domain, preventing multimerization and modulating synaptic signaling pathways. By linking mGluRs to intracellular calcium channels like the inositol triphosphate receptors (IP3R), Homer proteins facilitate the regulation of intracellular calcium dynamics, which is vital for numerous neuronal functions. Understanding the function and regulation of Homer proteins is important for research into neurological disorders and synaptic dysfunctions. Homer (D-3) monoclonal antibody is available in both unconjugated and conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, making it versatile for different experimental needs.

Does this antibody detect Homer1, Homer2, and Homer3 as well?

Thank you for your question. This mouse monoclonal antibody, Homer (D-3): sc-17842, is raised against Homer-1 of human origin (UniProt accession number: Q86YM7). The exact epitope recognized by this monoclonal antibody has not been determined, but based on sequence analysis there is approximately a 50% chance that this antibody may also cross-react with Homer-2 and Homer-3. This cross-reactivity has not yet been confirmed experimentally.
