    SEE ALSO...

HNP Antibody (H-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects HNP protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). HNP Antibody (H-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HNP protein, part of the defensin family, plays a crucial role in innate immune response, particularly in host defense against microbial infections. Defensins, including human neutrophil peptides (HNPs), are small cationic peptides produced predominantly by neutrophils and are essential for the body′s first line of defense. HNP-1, HNP-2, and HNP-3, which are secreted by neutrophils, constitute approximately 30% of total granule protein in these immune cells, highlighting their significance in combating pathogens. HNP-1 protein is encoded by the α1-defensin (DEFA1) gene, while HNP-3 is encoded by the α3-defensin (DEFA3) gene, and HNP-2 is a mature cleavage product of both precursor proteins. The presence of these peptides in neutrophil granules and their secretion during immune responses underscores their importance in phagocyte-mediated host defense, making anti-HNP antibody (H-2) a valuable tool for researchers studying immune responses and related pathologies.

Can HNP (H-2): sc-390796 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: TinTin
Thank you for your question. Yes, HNP (H-2): sc-390796 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 3 out of 5 by from Good on cells, not enought in FFPEThe antibody gives a signal not very cool in IHC on bone marrow biopsies (lot of background). The expression is good in ICC on alcool fixed cells. The best dilution is 1:300 using as antigen retrival the EDTA buffer pH 8.0 for IHC on FFPE and 1:100 for ICC.
