HMG-I/HMG-Y Antibody (D-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects HMG-I/HMG-Y protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). HMG-I/HMG-Y (D-12) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The high mobility group (HMG) I (Y) chromatin proteins play a crucial role in regulating gene expression by binding to the minor groove of AT-rich DNA sequences, which leads to significant alterations in DNA architecture, such as bending and unwinding of the helix. This structural modification is essential for facilitating the binding of transcription factors, as HMG proteins work synergistically with Oct-2, members of the NFkB family, ATF-2, and c-Jun to enhance transcriptional activation. Additionally, the phosphorylation of HMG proteins is vital for stimulating their transcriptional activity, highlighting the importance of post-translational modifications in their function. Human HMG I (Y) contains two distinct DNA-binding domains known as HMG boxes, which allow binding to single-stranded DNA while inducing conformational changes in double-stranded DNA, thereby influencing various cellular processes.
