HMG-1 Antibody (HAP46.5) is a mouse monoclonal IgG1 kappa light chain antibody that detects HMG-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-HMG-1 antibody (HAP46.5) is available as a non-conjugated format. The high mobility group (HMG) proteins, including HMG-1, play a crucial role in the regulation of gene expression and chromatin structure. Specifically, HMG-1 interacts with various transcription factors, such as Oct-2 and members of the NFκB family, enhancing their ability to activate transcription. This interaction is vital for cellular responses to stress and inflammation, as HMG-1 can act as a signaling molecule that influences the immune response. HMG-1 is predominantly located in the nucleus, where binding to DNA induces structural changes that facilitate the accessibility of transcription machinery to specific genes. The presence of two DNA-binding domains, known as HMG boxes, allows HMG-1 to bind to both single-stranded and double-stranded DNA, although primarily inducing conformational changes in double-stranded DNA. Understanding HMG-1 function and interactions is essential for elucidating its role in various biological processes, including development, differentiation, and cellular response to injury.

感谢您的提问。HMG-1/HMGB1 (HAP46.5): sc-56698 还没有经过CoIP实验的验证,因此,目前不推荐应用于CoIP. Thank you for your inquiry. We have not yet tested HMG-1/HMGB1 (HAP46.5): sc-56698 on CoIP. As a result, CoIP is not currently a recommended application for this antibody.

Does this antibody works well in Co-Immunoprecipitation experiments? What about immunocytochemistry?

Thank you for your question. We have not tested the HMG-1 Antibody (HAP46.5): sc-56698 for co-immunoprecipitation experiments, nor are we aware of anyone citing the antibody for this research application. The antibody is recommended and warrantied for immunoprecipitation of the HMG-1 antigen. We do not currently recommend this antibody for immunocytochemistry (or IF). However, there is a publication citing use of the antibody for this research application, see PMID: 24091659.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using withHMG-1 (HAP46.5): sc-56698?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. Is this antibody reactive with avian samples?

Yes, this antibody is expected to pick up avian HMG-1. However, we recomment to contact our Technical Service Department and we would be happy to perform sequence analysis and review our data to answer any questions you may have about additional species reactivity.
