HLA-A2 Antibody (BB7.2) is a mouse monoclonal IgG2b kappa light chain antibody that detects the HLA-A2 protein of human origin by flow cytometry (FCM). Anti-HLA-A2 antibody (BB7.2) is available in both non-conjugated and various conjugated forms, including phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. Human leukocyte antigen A2 (HLA-A2) plays a crucial role as a class I histocompatibility molecule, integral to the immune response by presenting peptides on the cell surface to cytotoxic T lymphocytes (CTLs). This function is vital for the recognition and elimination of infected or malignant cells, making HLA-A2 a key player in immune surveillance. Notably, HLA-A2 is implicated in autoimmune thyroid dysfunction in patients undergoing interferon-alpha therapy for chronic hepatitis C, highlighting HLA-A2′s significance in both immune response and disease pathology. Furthermore, research has shown that HLA-A2 antigens are downregulated in response to HIV, particularly in primary T cells, which can lead to evasion of the immune response. Such downregulation is also observed in various cell lines, including β and astrocytoma cells, indicating a broader impact on immune evasion strategies. Modifications to the heavy chain of HLA-A2 can enhance the presentation of specific HIV-1 epitopes, thereby improving the recognition and lysis of infected cells by HLA-A2 restricted CD8+ CTLs. Anti-HLA-A2 antibody (BB7.2) serves as an essential tool for researchers studying the complexities of immune responses, particularly in the context of viral infections and autoimmune diseases.

What negative control should be used when using the conjugated version of HLA-A2 (BB7.2): sc-32236 monoclonal antibody in flow cytometry?

Thank you for your inquiry. We recommend using the isotype specific normal mouse IgG conjugated to the same fluorophore. For example, if you are using sc-32236 PE, then you would want to use normal mouse IgG2b PE, sc-2868.
