HIP1 Antibody (4B10) is a mouse monoclonal IgG1 kappa light chain antibody that detects HIP1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. HIP1 (4B10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HIP1, known as huntingtin-interacting protein 1, plays a critical role in cellular processes, particularly in Huntington′s disease, where interaction with huntingtin is essential for maintaining membrane-cytoskeletal integrity in the brain. HIP1 is a membrane-associated protein that binds specifically to the N-terminus of huntingtin, with expression observed in various brain regions at low levels. The structural integrity of HIP1 is vital for function, containing an endocytic multidomain structure with a C-terminal actin-binding domain, a central coiled-coil region, and an N-terminal ENTH domain, which are all important for vesicle trafficking. Notably, interaction between huntingtin and HIP1 is inversely correlated with the length of the polyglutamine tract in huntingtin, suggesting that alterations in this interaction may contribute to the pathophysiology of Huntington′s disease. Additionally, HIP12, a non-proapoptotic member of the HIP gene family, shares a similar subcellular distribution pattern with HIP1 but does not directly interact with huntingtin, although HIP12 can interact with HIP1, highlighting the complex interplay of these proteins in neuronal function.

Thank you for your question. Yes, HIP1 (4B10): sc-47754 has been validated for use in IHC with paraffin-embedded sections.
