Hic-5 Antibody (H-1) is a mouse monoclonal IgG1 antibody that detects Hic-5 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Hic-5, also known as transforming factor beta 1 induced transcript 1, plays a crucial role in cellular signaling and adhesion through regulation of focal adhesion dynamics. Hic-5 contains four LIM motifs and seven zinc finger domains, which facilitate localization to the nuclear matrix and focal adhesion complexes. The LIM domains are essential for mediating interactions with focal adhesions, allowing anti-Hic-5 antibody (H-1) to inhibit integrin-mediated cell spreading on fibronectin by competing with paxillin for binding to focal adhesion kinase, thus preventing downstream signal transduction. This regulatory function is particularly important as increased expression of Hic-5 can lead to cellular senescence in developing fibroblasts, while during myogenesis, Hic-5 blocks differentiation and induces apoptosis in developing myoblasts. The gene encoding human Hic-5 is located on chromosome 16, highlighting Hic-5′s significance in various cellular processes and potential implications in developmental biology and disease.
