
Hic-5 Antibody (F-6) is a mouse monoclonal IgG2b antibody that detects hydrogen-peroxide inducible clone 5 (Hic-5) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Hic-5 is a crucial member of the LIM family, characterized by four LIM motifs and seven zinc finger domains, which play a significant role in mediating protein-protein interactions within the cell. Hic-5 (F-6) antibody recognizes a protein primarily localized to the nuclear matrix and focal adhesion complexes, where Hic-5 is essential for regulating cellular adhesion and signaling pathways. Notably, Hic-5 inhibits integrin-mediated cell spreading on fibronectin by competing with paxillin for binding to focal adhesion kinase, thereby disrupting downstream signaling cascades vital for cell migration and proliferation. Hic-5 involvement extends to cellular senescence, particularly in developing fibroblasts, where increased expression can lead to growth arrest. During myogenesis, Hic-5 expression blocks differentiation and induces apoptosis in developing myoblasts, highlighting a regulatory role in muscle development. The gene encoding human Hic-5 is located on chromosome 16, underscoring evolutionary significance and potential implications in various biological processes.
