HES1 Antibody (E-5) is a mouse monoclonal IgG2b kappa light chain antibody that detects HES1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). HES1 (E-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HES1 plays a crucial role in the regulation of myogenesis and neurogenesis, functioning as a transcriptional repressor within the Notch signaling pathway. This pathway is essential for proper cell differentiation and development, particularly during embryogenesis, where anti-HES1 antibody (E-5) helps to control segmentation and neural development. The HES gene family, which includes HES1, is homologous to the Drosophila Hairy and enhancer of split genes and encodes basic helix-loop-helix transcription factors that interact with the TLE proteins, the mammalian counterparts of groucho. This interaction is mediated by the carboxy-terminal WRPW motif of the HES proteins, allowing the HES/TLE complex to bind directly to DNA and regulate gene expression by repressing activator proteins. Notably, HES1 and HES5 preferentially bind to the N box (CACNAG) rather than the E box (CANNTG), highlighting their specific regulatory roles in various tissues. HES1 and HES2 are expressed in a range of adult and embryonic tissues, underscoring their importance in developmental processes and potential implications in various diseases.

Thank you for your question. While the regular version of this antibody is supplied as 200 µg in 1 ml PBS with gelatin and sodium azide, the X-formulated version is 200 µg in 0.1 ml with only sodium azide. Can the blocking peptide sc-166410 P be used in Western Blot as a positive control?
