HERG Antibody (F-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects HERG protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-HERG antibody (F-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The human ether-a-go-go related gene (HERG) encodes the pore-forming α subunit of the delayed rectifier potassium channel I_Kr, which plays a crucial role in cardiac repolarization. HERG protein is characterized by six transmembrane α-helices and a reentrant "pore-loop" that is essential for ion transport function. Notably, two N-terminal splice variants of HERG, isoform 1α and isoform 1β, exhibit different functional properties; isoform 1β, which lacks the PAS motif, deactivates more rapidly than isoform 1α, influencing channel kinetics. HERG expression is predominantly found in the heart, particularly in ventricles, where HERG is vital for maintaining electrical stability of cardiac cells. Mutations in HERG can lead to significant cardiac arrhythmias, including inherited long QT syndrome, by increasing beat-to-beat variability and early after depolarization, which can trigger premature heartbeats. Understanding HERG function and regulation is essential for elucidating mechanisms underlying cardiac health and disease.
