Hemoglobin α Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects the Hemoglobin alpha protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Hemoglobin α antibody (D-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Hemoglobin, a crucial protein in the body, is responsible for transporting oxygen from the lungs to tissues and facilitating the return of carbon dioxide from tissues back to the lungs. Hemoglobin′s structure consists of two alpha and two beta chains that form a tetramer, with each chain binding to a heme group that contains iron. This arrangement allows hemoglobin to undergo conformational changes that significantly affect oxygen-binding affinity. The α1-β2 cleavage plane plays a critical role in this process, as the interaction between the chains can either enhance or reduce oxygen binding, depending on whether the structure is in a relaxed or tense state. The majority of circulating hemoglobin is Hb A, which is composed of two alpha and two beta chains, while smaller amounts consist of Hb A-2 and fetal hemoglobin, highlighting Hemoglobin alpha′s importance in overall oxygen transport and delivery in the human body.

What Is the Manufacturing Expiry date for this item? Can I get a COA for the same?

Thank you for your question. To request a manufacture date please email the lot number to scbt@scbt.com . We do not provide an expiration date but we do have a 1 year warranty from date of purchase. Would like to know if there is any data regarding how does this antibody works with mouse-origin HBA and what kind of dilution could be suggested when using it to detect low abundant HBA (non-erythroid origin cells)

Thank you for your question. We do not recommend Hemoglobin α Antibody (D-4): sc-514378 for the detection of Hemoglobin α in murine samples. However, there is a publication that successfully used this antibody with murine samples in western blotting (PMID 29500821). The recommended starting dilution for human samples is 1:100 with a dilution range of 1:100-1:1000. For further information please contact technical service.

I want to run a Western blot using this Hemoglobin alpha monoclonal antibody using K-562 whole cell lysate. How much should I load on my gel?

Rated 5 out of 5 by from Worked wellWe used this antibody for WB of K562 cells. Loaded 20 ug protein, used primary Ab at 1/200 dilution, secondary at 1/1000 (anti-mouse-AP). Observed chemiluminescence band with lots of background (Immun-Star substrate), and a very clean band, no background using chromogenic substrate (BCIP/NBT).
