HAS2 Antibody (A-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects HAS2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). HAS2 Antibody (A-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HAS2, a member of the hyaluronan synthase family, plays a crucial role in synthesizing hyaluronic acid (HA), a vital component of the extracellular matrix that contributes to tissue hydration, elasticity, and cellular signaling. HA is a high molecular weight linear polysaccharide composed of alternating glucuronic acid and N-acetylglucosamine residues, linked by β-1,3 and β-1,4 glycosidic bonds. HAS2 synthesis of HA is particularly important in chondrocytes and cartilage, where HA supports the structural integrity and function of these tissues. HAS2 expression is notably upregulated in growing cells, indicating an essential role in development and tissue repair. The human HAS2 gene is located on chromosome 8q24.12, and distinct expression patterns during development highlight functional significance in various physiological processes.

Can HAS2 (A-7): sc-514737 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, HAS2 (A-7): sc-514737 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Hallo, wie wurden die Zellen für den WB lysiert (welcher Lysepuffer, Welche Aufbereitung)? Wurden die Proben vor der Elektrophorese reduziert? Wieviel Protein wurde je Spur aufgetragen?

We recommend to load 40–60 µg whole cell lysate per lane according to our protocol for Western Blotting. Rated 5 out of 5 by from clean antibody for WBThis antibody worked really well for my WB experiment. I used it for MDA-MB231 cell lysates, it just picked only one crisp band exactly at 63kDa, as suggested by data sheet. I used 1:800 dilution. Over all the blot was clean and there was no non-specific bands either.
