SEE ALSO...

GW182 Antibody (A-8) is a mouse monoclonal IgG2a antibody that detects GW182 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). GW182 plays a pivotal role in the formation and function of GW bodies (GWBs), which are essential for the regulation of mRNA stability and degradation. These structures serve as critical hubs for microRNA (miRNA) activity, and proper functioning of GW182 is vital for maintaining GWB integrity. Disruption of GW182 can lead to impaired silencing of small interfering RNAs (siRNAs) and miRNAs, which may contribute to various pathological conditions. Notably, GW182 interacts with endogenous argonaute-2 (Ago2), a key player in the RNA-induced silencing complex, further underscoring GW182′s importance in gene regulation. The presence of autoantibodies against GW182 has been observed in patients with autoimmune diseases such as ataxia, Sjögren′s syndrome, and sensor neuropathy, highlighting GW182′s potential as a biomarker for these conditions. Anti-GW182 antibody (A-8) serves as an invaluable tool for researchers investigating the intricate mechanisms of RNA regulation and the role of GWBs in cellular processes.

I am using GW182 (A-8): sc-377006 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Asked by: Randy McDonald
We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
