GR Antibody (G-5) is a mouse monoclonal IgG2b kappa light chain antibody that detects GR (glucocorticoid receptor) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). GR (G-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GR (glucocorticoid receptor) is a critical transcription factor that plays a vital role in mediating the physiological effects of glucocorticoids, which are steroid hormones involved in numerous biological processes such as metabolism, immune response, and stress regulation. GR (glucocorticoid receptor) is predominantly located in the cytoplasm, where GR exists in an inactive form bound to heat shock proteins, particularly HSP90. Upon binding to glucocorticoids, GR (glucocorticoid receptor) translocates to the nucleus, where GR regulates gene expression by either inducing or repressing target genes. This translocation allows GR (glucocorticoid receptor) to interact with specific DNA sequences and recruit co-regulators that modulate transcription. GR (glucocorticoid receptor)′s ability to influence gene expression is essential for maintaining homeostasis and responding to environmental changes, making GR a significant target for therapeutic interventions in conditions such as inflammation, autoimmune diseases, and cancer.

I have delta GR in which we have only the c-terminus of GR from 506 aa to the 794 aa so which antibody would be suitable for this deleted version of GR?

Setting: Western Blot 20 mcg protein of tissue lysate (mouse tissue); antibody diluted 1:1000 in 5%BSA. Problem: I have a good band at 95 kDa , but also a clear (unexpected) band around 50kDa. What could this be?

Does GR antibody (G-5): sc-393232 work in ChIP assay for rat muscle?

Thank you for your question. This antibody has been published previously for ChIP applications (PMID: 30994266 and PMID: 30698747). This antibody is reactive with mouse, rat and human. We do not have any data specifically in rat muscle.

How well does the antibody work with rat brain tissue for western blotting, and what would be the optimum concentration?

Thank you for your question. We have not yet tested this antibody in rat brain tissue. We do have data in mouse brain tissue extract. The primary antibody was used at the starting dilution of 1:200, and loading 50ug tissue extract.

Is the GR ANTIBODY (G-5) X sc-393232X the same as the GR ANTIBODY (G-5) sc-393232 but just at a higher concentration?

Thank you for your question. Yes, sc-393232 X is provided at a 10x higher concentration. It is provided as 200µg in 0.1ml while the standard product is provided as 200µg in 1ml.

Hi, please let me know what concentration of the  GR Antibody (G-5): sc-393232 would be optimal for  Flow Cytometry.  Thank you.

Thank you for your question. We have not yet tested this mouse monoclonal, GR Antibody (G-5): sc-393232, for Flow Cytometry so this application is not recommended at this time. We do offer this antibody directly conjugated to a variety of fluorochromes, including: phycoerythrin (sc-393232 PE), fluorescein (sc-393232 FITC), Alexa Fluor® 488 (sc-393232 AF488) or Alexa Fluor® 647 (sc-393232 AF647), 200 µg/ml, for IF, IHC(P) and FCM. If you purchase one of the direct conjugates, we can extend our product warranty to cover the application of Flow Cytometry. A link to our recommended protocol for FCM is below. We ran a dilution series in mouse brain on free floating 50-micron sections for the following primary antibody dilution amounts: 1:50, 1:100, and 1:200 (+ a no primary control). There was minimal difference between these concentrations and no staining present in the no primary control so we plan to proceed by using it at 1:100.

Rated 4 out of 5 by from Glucocorticoid receptor antibodyGood staining of mice hypothalamus. Sample: mouse cortex fixed with 4% PFA for 16hr at 4℃, followed by sucrose infiltration and OCT embedding. Staining: floatation staining for 48hr at 4℃ (1:300 or 1:500, 300 was better), followed by 2nd antibody staining. FYI, I have not tried shorter staining such as 1hr at RT or overnight at 4℃.

Rated 5 out of 5 by from PERFECT CHOICEThis antibody works well. Used 1:500 in tbs 0,1%tween to detect GR in different cell line extract (45gs of total proteins; MCF-7 breast cancer cells and KP2 and SUIT2 pancreatic cancer cells), detect only one specific band at 95 kDa. I’ll buy this antibody.
