GR Antibody (FiGR) is a mouse monoclonal IgG2b kappa light chain antibody that detects glucocorticoid receptor (GR) protein of mouse, rat, and human origin by western blotting (WB) and immunoprecipitation (IP). GR monoclonal antibody (FiGR) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Glucocorticoid receptor is a critical transcription factor that plays a vital role in mediating physiological effects of glucocorticoids, which are steroid hormones involved in numerous biological processes such as metabolism, immune response, and stress regulation. GR protein exists in two main isoforms, with GR α being most prevalent and functionally significant. GR α remains inactive in cytoplasm until bound by glucocorticoids, leading to translocation into nucleus. GR α binds to specific DNA sequences in nucleus, regulating transcription of target genes essential for processes like apoptosis, cell growth, and differentiation. GR interacts with various co-regulators and transcription factors, including AP-1 complex, which is crucial for modulating gene expression in response to hormonal signals. Understanding GR function and dynamics is essential for elucidating roles in health and disease, particularly in conditions related to inflammation, metabolic disorders, and cancer.

When should the TransCruz reagent sc-12763 X be used instead of the standard form sc-12763?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.

Detected GR at expected size in 15 ug total proteine lysate, wet transfer one hour at 450 mAmps on PVDF membrane, overnight blocking with 5% nonfat dry milk in PBS, one hour incubation at RT with 1:500 anti GR in 5% PBS with 0,2%Tween followed by one hour incubation with 1:10.000 anti mouse (sc-358919) HRP conjugated antibody and detection with ECL prime substrate followed by short film exposure (under 1 minute) and automatic processing.
