
Geminin Antibody (A-3) is a mouse monoclonal IgG1 antibody that detects geminin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Geminin is a crucial nuclear protein that plays a vital role in regulating DNA replication initiation during the cell cycle. Anti-geminin antibody (A-3) functions by inhibiting the assembly of minichromosome maintenance (MCM) proteins with chromatin, thereby preventing the formation of the prereplication complex essential for DNA replication. Geminin levels fluctuate throughout the cell cycle, being lowest during the G1 phase and rising during the S, G2, and M phases, before decreasing again during mitosis. This cyclical expression is critical, as geminin degradation during mitosis, mediated by the anaphase-promoting complex (APC), allows for DNA replication initiation in the subsequent G1 phase. APC specifically targets proteins with a destruction box motif, including B-type cyclins, for degradation via ubiquitin-mediated proteolysis, highlighting geminin′s regulation importance in maintaining genomic stability and preventing untimely DNA replication.
