GADD 45α Antibody (4T-27) is a mouse monoclonal IgG1 kappa light chain antibody that detects GADD 45α in mouse, rat, and human samples through western blotting, immunoprecipitation, and enzyme-linked immunosorbent assay. GADD 45α monoclonal antibody (4T-27) is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. GADD 45α plays a crucial role in cellular response to DNA damage, particularly in regulating cell cycle checkpoints. When DNA damage occurs, cell cycle can be halted at G1 and G2 checkpoints, allowing for necessary repair processes before proceeding to DNA synthesis and mitosis. Tumor suppressor protein p53 enhances GADD 45α and p21 expression, with p21 inhibiting cyclin-dependent kinase (Cdk) activity to suppress cell proliferation. GADD 45α interacts with Cdks and proliferating cell nuclear antigen (PCNA), a key player in DNA replication and repair. GADD 45α enhances DNA excision repair in vitro and prevents cells from entering S phase, suggesting a critical link between p53-mediated cell cycle regulation and DNA repair mechanisms.

To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody.
