GADD 153 Antibody (H-5) is a mouse monoclonal IgG3 kappa light chain antibody engineered to detect GADD 153, also known as CHOP or DDIT3, across multiple species including mouse, rat, human, equine, canine, bovine, and porcine using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) techniques. GADD 153 monoclonal antibody (H-5) is raised against a peptide mapping at the C-terminus of human GADD153, ensuring precise and reliable binding for various experimental applications. GADD 153 (H-5) antibody recognizes a critical C/EBP-related nuclear transcription factor that plays a pivotal role in cellular stress responses, particularly in endoplasmic reticulum (ER) stress and unfolded protein response (UPR). Structurally, GADD 153 contains a basic leucine zipper (bZIP) domain that facilitates dimerization with other transcription factors such as ATF4 and members of the C/EBP family, enabling binding to specific DNA sequences and regulation of gene expression. These interactions are essential for promoting apoptosis and cell cycle arrest under prolonged stress conditions by modulating the expression of genes involved in these pathways. GADD 153 also interacts with proteins like Bcl-2 and Bim, which are key regulators of apoptotic machinery, thereby integrating stress signals to execute programmed cell death when necessary. Anti-GADD 153 antibody (H-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants, to accommodate diverse experimental requirements.

Hi, for IHC use, what is the suggested pH for antigen retrieval pH? Citrate buffer pH 6.0 or TE buffer pH 9.0? Thanks!

Asked by: JessyQ
Thank you for your question. We would suggest using sodium citrate buffer pH 6 for antigen retrieval.

What is the protocol for the negative control blocking peptide sc-166682 P?

Asked by: AbPolly
Thank you for your question. The blocking peptide is intended for use by preincubating with the primary antibody to block the specific reactivity. The details of the protocol can be found on our website here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization

As "GADD153(H-5) :sc-166682" antibody is labeled as "IgG2b (kappa light chain)", could I use anti-mouse IgG secondary antibody? Or I need to use special secondary antibody?

Asked by: huihui
Thank you for your question. Yes, you can use any anti-mouse IgG secondary antibody for detection of our primary antibody, GADD153 (H-5): sc-166682. This primary antibody has an isotype of IgG2b (kappa light chain). An isotype-specific secondary antibody may be used, but it is not required.
