Fra-1 Antibody (D-3) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 1-50 of human Fra-1, a crucial transcription factor in the Fos protein family. Anti-Fra-1 antibody (D-3) recognizes an epitope within this N-terminal region and demonstrates broad species cross-reactivity, detecting Fra-1 in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry on paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. As a nuclear phosphoprotein also known as Fos-related antigen 1 or FOSL1, Fra-1 functions through sophisticated protein interactions. Through its basic leucine zipper (bZIP) domain, Fra-1 forms heterodimers with Jun family proteins (c-Jun, JunB, and JunD) to create the activator protein-1 (AP-1) complex, enabling specific DNA binding and transcriptional regulation of genes controlling cell proliferation, differentiation, and survival. Fra-1′s regulatory reach extends through interactions with ATF/CREB family members, expanding Fra-1′s influence over various cellular processes. These protein partnerships prove essential for Fra-1′s ability to respond to extracellular signals and coordinate complex gene expression programs. When Fra-1 regulation becomes disrupted, particularly through overexpression, the resulting dysregulation can contribute to tumorigenesis and cancer progression. Through these diverse cellular functions, Fra-1 (D-3) antibody provides researchers with a valuable tool for investigating both normal physiology and disease mechanisms.

Thank you for your question. For Western Blotting experiments with this antibody we recommend a starting dilution of 1:100 and a dilution range of 1:100-1:1000.

Hello, I would like to use this antibody for ChIP. What dilution would you recommend? Do you have any validation data on the ChIP performance on this antibody?

Thank you for your question. This antibody has not yet been tested for ChIP, and we are not yet aware of any product citations using this particular antibody for any ChIP applications. We do extend our warranty to cover that research application for our TransCruz reagent for Gel Supershift and ChIP applications (sc-376148 X, 200 µg/0.1 ml). We currently offer a different antibody that has been published for use in ChIP applications. Please refer to our Fra-1 Antibody (C-12): sc-28310. And PMID's: 22198213 and 21088499.

Welche Proteinmenge eignet sich am besten für einen Western Blot mit dem Fra-1 Antikörper?

Vielen Dank für Ihre Frage. Die optimale Proteinmenge ist abhängig von dem verwendeten Sample. Generell empfehlen wir pro Bahn bei der Elektrophorese eine Proteinmenge von 40–60 µg bei Zelllysaten, 10–20 µg bei Kernextrakten, 5-10 µg bei transfizierten Zelllysaten, oder 10–20 ng bei aufgereinigtem Protein. What amount of protein is ideal for a western blot with Fra-1 Antibody (D-3): sc-376148? Thank you for your question. The ideal amount of protein is dependent on the sample that is being tested. We generally recommend 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.

I would like to use Fra-1 Antibody (D-3): sc-376148 for double staining with another mouse monoclonal primary antibody. Is this possible?
