Fra-1 Antibody (C-12) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 1-50 of human Fra-1, a crucial transcription factor in the Fos protein family. Anti-Fra-1 antibody (C-12) recognizes an epitope within this N-terminal region and demonstrates broad species cross-reactivity, detecting Fra-1 in mouse, rat, and human samples through Western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry on paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. As a nuclear phosphoprotein also known as Fos-related antigen 1, FRA1, or FOSL1, Fra-1 functions through a sophisticated mechanism involving its basic leucine zipper (bZIP) domain, which enables heterodimerization with Jun family proteins to form the activator protein-1 (AP-1) complex. This complex binds to specific DNA response elements, regulating genes involved in cell proliferation, differentiation, and survival. Fra-1′s activity undergoes precise control through post-translational modifications, particularly phosphorylation by kinases such as ERK, which stabilizes Fra-1 by preventing its degradation via the ubiquitin-proteasome pathway. This stabilization mechanism takes on particular significance in cancer biology, where Fra-1 overexpression and accumulation can drive tumor progression and metastasis. Through these diverse cellular functions and regulatory mechanisms, Fra-1 (C-12) antibody provides researchers with a valuable tool for investigating both normal physiology and disease pathways.
