FOXM1 Antibody (G-5) is a mouse monoclonal IgG2b kappa light chain antibody engineered to detect FOXM1 across mouse, rat, and human species through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). FOXM1 G-5 is mouse monoclonal IgG2b. I would like to know if it can be used with conventional secondary anti-mouse IgG reagents.

Thank you for your question. Yes, you can use a conventional anti-mouse IgG secondary antibody for detection of this primary antibody. For more specific results, we do recommend using an isotype-specific secondary antibody (anti-mouse IgG2b), or our mouse IgGκ binding protein, e.g., m-IgGκ BP-HRP: sc-516102.

FOXM1, also known as FKHL16, MPP2 or Trident, is primarily expressed in proliferating cells. The gene encoding human FOXM1 maps to chromosome 12p13.33. The transcription element that restricts FOXM1 expression to prolif- erating cells is located 300 bp upstream of the start codon. FOXM1 is most abundant in thymus, testis, small intestine and colon. Alternative splicing generates FOXM1A and FOXM1B isoforms that contain PEST regions involved in rapid protein degradation. A decrease in FOXM1 expression is associated with age-related defects in cellular proliferation. Conversely, an increase in FOXM1B expression in the livers of older transgenic mice restore hepatocyte DNA replication rates to the higher rate present in young livers. FOXM1B activates the transcription of cyclin B1, cyclin D1 and Cdc25B

Rated 1 out of 5 by from Not sure if observed band is FOXM1Like the previous comments below, our group has also confirmed that this antibody may not be recognizing FOXM1. We observe a strong clear band at around 125-130 kDa with this antibody in our cell lines. However, upon using both siRNA and shRNA approaches with 3 different species, we could not observe a clear knockdown of FOXM1 with this antibody. In contrast, FOXM1 (A-11) sc-271746 showed knockdown of FOXM1 protein. Additionally, while this antibody can recognize overexpressed FOXM1, the exogenous FOXM1 appears as a second at around 110 kDa. The FOXM1 (A-11) antibody only recognizes a single band with much greater intensity. The FOXM1 (A-11) antibody also gave consistent results with CS #D12D5 and an Abcam antibody.

Rated 3 out of 5 by from Gives good band, but uncertain about specificityThe mouse G-5 antibody appears to detect primarily nuclear FOXM1; whereas the SC-500 rabbit antibody detects mainly cytoplasmic (and to some extent nuclear). The expression pattern in western blots, ICC, and IHC have confirmed this (images attached). Possibly due to the G-5 antibody unable to detect FOXM1 in cytoplasm if dimerized? It seems to detect FOXM1 differently in regards to localization; however, the MW appears to be correct. I still haven't quite figured this one out. In any case, I used the antibody at a 1:1000 dilution and secondary at 1:2500. I've noticed most SC antibodies work well at 1:250/500 primary and secondary at 1:2500. It has worked well with overexpression (human FOXM1b isoform) as well as siRNA knockdown in rat cells.
