FAT10 Antibody (G-5) is a mouse monoclonal IgG1 antibody that detects FAT10 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). FAT10, also known as Ubiquitin D or Diubiquitin, is a 165 amino acid protein encoded within the major histocompatibility complex (MHC) and features two domains that exhibit significant homology with ubiquitin. Each of these domains contains two cysteine residues and a free C-terminal diglycine motif, which is essential for the formation of FAT10 conjugates. FAT10 is notably inducible by interferon-γ and tumor necrosis factor alpha (TNFα), highlighting FAT10′s role in immune responses. FAT10 is crucial for various cellular processes, including antigen presentation, cytokine response, apoptosis, and mitosis, and FAT10 interacts with MAD2, a key component of the spindle checkpoint, thereby influencing cell cycle regulation. Expression of FAT10 mRNA is primarily observed in certain dendritic cells and lymphoblastoid lines, particularly following interferon-γ induction. Furthermore, the human FAT10 gene, designated UBD, is located on chromosome 6p21.3 and has been found to be overexpressed in tumors of various epithelial cancers, making FAT10 a significant marker for cancer research and potential therapeutic targeting.
