FAT10 Antibody (A-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects FAT10 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-FAT10 antibody (A-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FAT10, also known as Ubiquitin D or Diubiquitin, is a 165 amino acid protein encoded within the major histocompatibility complex (MHC) and features two domains that exhibit significant homology with ubiquitin. Each domain contains two cysteine residues and a free C-terminal diglycine motif, which is essential for the formation of FAT10 conjugates. FAT10 is notably inducible by interferon-γ and tumor necrosis factor alpha (TNFα), highlighting its role in immune responses. FAT10 is crucial for various cellular processes, including antigen presentation, cytokine response, apoptosis, and mitosis, and interacts with MAD2, a key component of the spindle checkpoint, thereby influencing cell cycle regulation. Additionally, FAT10 may play a role in regulating cell growth during the activation and development of dendritic cells and B cells. FAT10 mRNA expression is primarily observed in certain dendritic cells and lymphoblastoid lines, particularly following interferon-γ induction. The human FAT10 gene, designated UBD, is located on chromosome 6p21.3 and has been found to be overexpressed in tumors of various epithelial cancers, making FAT10 a significant target for research in cancer biology and immunology.
