FANCL Antibody (B-11) is a mouse monoclonal IgG2b kappa light chain antibody that detects FANCL in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). FANCL (B-11) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. FANCL plays a crucial role in the Fanconi anemia (FA) pathway, which is essential for maintaining genomic stability. Defects in FANCL can lead to Fanconi anemia, an autosomal recessive disorder characterized by bone marrow failure, developmental abnormalities, and increased susceptibility to chromosomal breakage. FANCL functions as a ligase, mediating the ubiquitination of FANCD2, a pivotal step in the DNA damage response. This ubiquitination is vital for the recruitment of repair proteins to sites of DNA damage, thereby facilitating the repair process. Additionally, phosphorylation of FANCL and other FA proteins is believed to be critical for their function within the FA pathway, highlighting the importance of post-translational modifications in regulating protein interactions and activities. FANCL may also be necessary for proper primordial germ cell proliferation during embryonic development, underscoring its significance in both DNA repair and developmental processes.

For FANCL western blot from whole cell lysate what dilution of antibody should be used? Also please tell how long it should be incubated. I used antibody at a dilution of 1:500 and incubated for 4 hours at RT. I am getting multiple specific bands.

Thank you for your question. We recommend using this antibody initially at the starting dilution of 1:200 (dilution range 1:100-1:1,000). We recommend incubating for 1 hour at room temperature in 5% nonfat milk + TBST. Please refer to our protocol online for more detailed information.

I am using FANCL (B-11): sc-137067 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
