ESD Antibody (D-4) is a mouse monoclonal IgG1 antibody that detects ESD (esterase D) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ESD is a 282 amino acid protein belonging to the esterase D family, primarily expressed in the placenta, kidney, liver, and erythrocytes, and is localized in the cytoplasm and cytoplasmic vesicles. One of the critical functions of ESD is its role in detoxifying formaldehyde, a harmful compound that can disrupt cellular processes. ESD achieves this by catalyzing the hydrolysis of S-formylglutathione, a product of formaldehyde oxidation by alcohol dehydrogenase 5, into the reduced forms of formic acid and glutathione, thereby mitigating potential toxicity. Additionally, ESD exhibits carboxylesterase activity, hydrolyzing various neutral ester substrates, and may function as a cysteine hydrolase, although ESD is susceptible to inactivation by thiol alkylating agents. Genetic polymorphisms in the ESD gene can lead to reduced enzymatic activity, which has been associated with an increased risk of conditions such as toxic liver cirrhosis, retinoblastoma, obesity, and autism. Anti-ESD antibody (D-4) is an essential tool for researchers investigating the biochemical pathways involving formaldehyde detoxification and the implications of ESD in various diseases.
