dUTPase Antibody (G-6) is a mouse monoclonal IgG2b antibody that detects dUTPase in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). dUTPase, also known as deoxyuridine 5′-triphosphate nucleotidohydrolase or DUT, plays a critical role in nucleotide metabolism as a preventive DNA repair enzyme. DUT-N represents the most abundant nuclear isoform, while DUT-M exists as a mitochondrial variant in certain tissues. Nuclear localization of dUTPase enables efficient hydrolysis of dUTP to dUMP and diphosphate in the presence of magnesium ions, maintaining low intracellular dUTP levels. This regulation prevents excessive uracil incorporation into DNA, which can trigger genomic instability and cell death, highlighting dUTPase′s essential role in cell viability and potential as a target for anticancer therapies. dUMP, produced through this hydrolysis reaction, serves as a precursor for thymidine nucleotides, which are indispensable for DNA replication, underscoring dUTPase′s importance in cellular proliferation and maintenance.
