DR5 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects DR5 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-DR5 antibody (D-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. DR5 (TNFRSF10B) plays a crucial role in mediating apoptosis through interaction with cytokine TRAIL (TNF-related apoptosis-inducing ligand). This interaction regulates programmed cell death, essential for maintaining cellular homeostasis and eliminating damaged or cancerous cells. DR5′s ability to initiate apoptosis makes DR5 a significant target in cancer therapy, as enhancing DR5 signaling pathway can promote death of tumor cells while sparing normal cells. DR5 belongs to tumor necrosis factor receptor superfamily, which includes receptors like FAS and TNF-R1, underscoring DR5′s relevance in immune responses and therapeutic interventions.

We recommend using DR5 (D-6): sc-166624 mouse monoclonal antibody at a starting dilution of 1:200 for Western blot (range of 1:100-1:1,000).

Is it antibody targeting the internal or external part of the receptor? According to RCSB the epitope mapping between amino acids 48-76 falls in the extracellular part of the receptor?

This antibody is specific for an epitope mapping between amino acids 48-76 within an internal region of DR5 of human origin and should detect the extracellular part of the DR5 (amino acids 56-444 are extracellular per accession#O14763). Rated 4 out of 5 by from Clear and good signalPrimary antibody DR5 (D-6) sc-166624 was used diluted 1:500 according to the manufacturer’s recommendations (for Western Blotting dilution range 1:100-1:1000), in Milk 5%. Membranes of PVDF have been incubated overnight at 4°C and then they have been probed for 1h at RT with secondary antibodies diluted 1:5000 in Milk 2,5%. We have tested this antibodiy on melanoma cell line A375 and A2058. In both cases we obtained a clear and quite good signal, so we can conclude that the antibody has worked very well.

Rated 1 out of 5 by from tested on rat cells (NRK52E)Tested in WB on NRK52E cell lysate + BJAB cell lysate (used as positive control) Expected MW at 48kDa We have observed 2 different bands in WB at 72kDa and 25kDa. Nothing at 48kda. Probably, it is possible that the antibody is not specific for rat cells. In any case, we have seen similar results also in BJAB cell lysate (no clear band at 48kDa).
