desmoplakin I/II Antibody (A-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects desmoplakin I/II protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). desmoplakin I/II (A-1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Desmoplakin serves as a crucial component of desmosomes, specialized cell adhesion structures providing mechanical strength to tissues, particularly in epidermis and cardiac muscle. Desmoplakin anchors intermediate filaments to desmosomal plaques, maintaining cell-cell adhesion integrity under stress. Desmoplakin interacts with plakophilins, including plakophilin 1, plakophilin 2, and plakophilin 3, facilitating recruitment of these proteins to desmosomes, essential for junction formation and stability. Desmoplakin′s structure includes a two-stranded coiled-coil domain, and alternative splicing produces two isoforms, desmoplakin I and II, differing by 600 amino acids, demonstrating complexity in desmosome assembly and function. Disruptions in desmosomal integrity can lead to various skin and cardiac diseases, including pemphigus vulgaris and arrhythmogenic right ventricular cardiomyopathy.
