DAP-5 Antibody (F-2) is a mouse monoclonal IgG2b kappa light chain antibody that detects DAP-5 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). DAP-5 (F-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Death-associated protein 5 (DAP-5), also known as p97 and NAT1, is a crucial member of the eukaryotic translation initiation factor-4G (eIF4G) family, playing a significant role in protein synthesis regulation. DAP-5 is ubiquitously expressed and highly conserved across species, which underscores its importance in cellular functions. When activated by FAS or p53, DAP-5 undergoes caspase cleavage at position 790, resulting in a C-terminal truncated form that can form complexes with eIF4A and eIF3. This truncated protein lacks the N-terminal region of eIF4G that binds to the cap-binding protein eIF4E, allowing DAP-5 to act as a general repressor of translation by forming inactive complexes. During apoptosis, the caspase-activated DAP-5 can mediate cap-independent translation from its own internal ribosome entry site, creating a positive feedback loop that sustains DAP-5 translation. DAP-5 is essential for cellular differentiation, as DAP-5 regulates specific gene expression pathways, highlighting its multifaceted role in both normal cellular processes and stress responses.

Thank you for your question. Yes, DAP-5 (F-2): sc-137011 has been validated for use in IHC with paraffin-embedded sections.
