DAP-3 Antibody (E-4) is a mouse monoclonal IgG1 antibody that detects DAP-3 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). DAP-3 is a crucial nucleotide-binding protein that plays a significant role in the regulation of programmed cell death, making DAP-3 essential for understanding apoptosis mechanisms. DAP-3 contains a potential P-loop motif and functions downstream of receptor signaling complexes, with pro-apoptotic effects being dependent on caspase activity. Notably, DAP-3 interacts with the glucocorticoid receptor (GR) primarily through its amino-terminal region, which acts in a dominant-negative manner to protect cells from undergoing apoptosis. This interaction highlights DAP-3′s importance in cellular survival and stress responses. DAP-3 is conserved across species at both functional and structural levels and is widely expressed in highly proliferative epithelial tissues, indicating DAP-3′s fundamental role in cellular processes. Interestingly, unlike many other proteins involved in apoptosis, DAP-3 maintains mitochondrial localization during cell death induction, underscoring DAP-3′s unique function in apoptosis regulation.
