Cryopyrin Antibody (H-8) is a mouse monoclonal IgG2b kappa light chain antibody engineered to detect human Cryopyrin protein, also known as NLRP3 or NALP3. Cryopyrin is an essential component of the NLRP3 inflammasome, a multi-protein complex located in the cytoplasm that plays a crucial role in the innate immune response by recognizing pathogenic microorganisms and cellular stress signals. Structurally, Cryopyrin comprises a central nucleotide-binding and oligomerization domain (NACHT), multiple leucine-rich repeats (LRRs) at the C-terminus, and an N-terminal pyrin domain (PYD), which facilitate interactions with other proteins. Cryopyrin interacts with apoptosis-associated specklike protein containing a CARD domain (ASC), which is vital for the recruitment and activation of caspase-1, leading to the processing and release of pro-inflammatory cytokines such as IL-1β and IL-18. Additionally, Cryopyrin interacts with NEK7, a serine/threonine kinase that is critical for NLRP3 inflammasome activation. Anti-Cryopyrin antibody (H-8) binds to an epitope between amino acids 200-221 of Cryopyrin, and is recommended for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications in human samples. Understanding Cryopyrin′s interactions and functions is essential, as mutations in the NLRP3 gene are associated with several autoinflammatory disorders, including familial cold autoinflammatory syndrome (FCAS), Muckle-Wells syndrome (MWS), and chronic infantile neurologic cutaneous and articular syndrome (CINCA).
