CREM Antibody (C-2) is a mouse monoclonal IgG2a kappa light chain antibody specifically developed to recognize the cAMP response element modulator (CREM). Anti-CREM antibody (C-2) targets the N-terminal region of CREM, encompassing amino acids 1-153, and is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) across mouse, rat, and human species. CREM is a vital transcription factor within the ATF/CREB family, playing a critical role in regulating gene expression in response to cAMP signaling pathways. The functionality of CREM is significantly influenced by post-translational modifications such as phosphorylation, which are essential for modulating transcriptional activity, stability, and interactions with other proteins. These modifications are crucial for CREM′s involvement in key biological processes, including spermatogenesis, where CREM activity is indispensable for male fertility, as evidenced by infertility in male mice lacking a functional CREM gene. Additionally, CREM interacts with tissue-specific partners like ACT (activator of CREM in the testis), whose activity is regulated by the testis-specific kinesin KIF17b, highlighting the importance of post-translational regulation in function. CREM′s ability to influence cellular differentiation, proliferation, and apoptosis underscores significance in maintaining cellular homeostasis and potential impact on various physiological and pathological conditions. CREM (C-2) antibody is also available in multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates, providing versatility for diverse experimental needs.

How can CREM (C-2): sc-390426 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to HRP (sc-390426 HRP), phycoerythrin (sc-390426 PE), fluoroscein (sc-390426 FITC) and AlexaFluors.

Does it also detect ICER (1pprox. 20kDa band) on a western blot?

Thank you for your question. We have not specifically tested this antibody to determine if it can also detect the isoform known as ICER. CREM Antibody (C-2) is a mouse monoclonal raised against amino acids 1-153 mapping at the N-terminus of CREM of mouse origin. The exact epitope recognized by the antibody has not been determined. There are many isoforms described for CREM, and most are modified within the range of the immunogen for this antibody. This would have to be determined experimentally.

Rated 4 out of 5 by from Recognizes all CREB family members in WBI overexpressed GFP-tagged ATF1, CREB and CREM in HEK293 cells. CREM C2 (dilution 1:200) recognized GFP-CREM, but was not specific to CREM in WB, as it recognised all the GFP-tagged CREB family members. Also recognizes endogenous levels of CREB in rat primary cortical neuron lysates.
