
CREB-1 Antibody (D-12) is a mouse monoclonal IgG2a kappa light chain antibody engineered to target amino acids 254-327 of human CREB-1, providing reliable detection of CREB-1A, CREB-1B, CREM, and ATF-1 isoforms across mouse, rat, human, and avian species, as well as additional species including equine, canine, bovine, and porcine, through western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. CREB-1, also called cAMP response element-binding protein 1, CREB, and CREB1, is a crucial transcription factor located primarily in the cell nucleus, where CREB-1 binds to cAMP response elements (CRE) in the promoters of target genes, thereby regulating essential cellular processes such as metabolism, neuronal plasticity, and cell survival. The functionality of CREB-1 is intricately modulated through interactions with several key proteins, including CREB-binding protein (CBP), which acts as a coactivator facilitating the assembly of the basal transcription complex upon CREB-1 phosphorylation at Serine 133 by protein kinase A. Additionally, CREB-1 interacts with the nuclear coactivator PGC-1α, playing a significant role in neoglucogenesis and energy metabolism, as well as with other regulatory proteins such as TORC (transducers of regulated CREB activity) that enhance transcriptional activity. These protein-protein interactions are essential for the precise regulation of gene expression in response to hormonal and stress signals, highlighting CREB-1′s involvement in critical pathways related to cellular adaptation and function. Furthermore, the dynamic interactions of CREB-1 with binding partners underscore CREB-1′s importance in the pathogenesis of type II diabetes and dilated cardiomyopathy, where dysregulation of these interactions can lead to metabolic and cardiovascular dysfunctions.
