SEE ALSO...

CREB-1 Antibody (D-4) is a mouse monoclonal IgG2a kappa light chain antibody designed to target amino acids 1-341 of human CREB-1, facilitating the detection of CREB-1A, CREB-1B, CREM, and ATF-1 isoforms across mouse, rat, and human species using western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA) applications. CREB-1, also referred to as CREB, CREB1, and cAMP response element-binding protein 1, is a vital transcription factor localized primarily in the cell nucleus, where CREB-1 binds to cAMP response elements (CRE) in the promoters of target genes, thereby regulating essential cellular processes such as metabolism, neuronal plasticity, and cell survival. A key aspect of CREB-1′s functionality involves post-translational modifications, particularly phosphorylation at Serine 133 by protein kinase A, which is crucial for activation and subsequent interaction with the CREB-binding protein (CBP). This phosphorylation event induces a conformational change in CREB-1, enabling the recruitment of CBP and the assembly of the basal transcription complex, thereby initiating the transcription of genes involved in cellular responses to hormonal and stress signals. Additionally, CREB-1 can undergo other post-translational modifications such as acetylation and ubiquitination, which regulate stability, DNA-binding affinity, and interaction with other coactivators, further fine-tuning gene expression. These modifications are essential for the precise control of CREB-1 activity, influencing processes like neoglucogenesis through interactions with the nuclear coactivator PGC-1 and playing significant roles in the pathogenesis of type II diabetes and dilated cardiomyopathy. anti-CREB-1 antibody (D-4) is an indispensable reagent for researchers exploring the complex regulatory mechanisms governed by CREB-1 and its impact on cellular function and disease progression.

Can CREB-1 (D-4): sc-374227 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Cweed
Thank you for your question. Yes, CREB-1 (D-4): sc-374227 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
