CNTF Antibody (H-8) is a mouse monoclonal IgG2a antibody that detects ciliary neurotrophic factor (CNTF) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CNTF is a crucial neuropoietic cytokine that promotes the survival and differentiation of various cell types, including sensory, sympathetic, and motor neurons, vital for maintaining nervous system integrity. CNTF (H-8) antibody targets proteins primarily located in the central nervous system and peripheral tissues, where CNTF exerts neuroprotective effects by binding to its receptor complex, including CNTFRα and the gp130/LIFRb heterodimer. This interaction activates downstream signaling pathways, particularly Stat1 and Stat3 transcription factors, leading to enhanced DNA synthesis and neuronal survival. CNTF has been implicated in amyotrophic lateral sclerosis (ALS), highlighting potential therapeutic applications. While mice lacking CNTF exhibit certain deficits, those with a homozygous null mutation in the CNTFRα gene experience severe motor neuron deficits and perinatal mortality, suggesting CNTFRα′s essential role in neonatal nervous system development and indicating the presence of additional CNTFR ligands critical during this developmental phase.
