Claspin Antibody (B-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects Claspin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Claspin Antibody (B-6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Claspin plays a critical role in the DNA damage response by acting as a replication checkpoint control protein, which is essential for maintaining genomic stability. Anti-Claspin antibody (B-6) regulates the interaction between checkpoint kinase 1 (Chk1) and the upstream regulatory kinase ATR, facilitating cell cycle arrest in response to DNA replication stress or damage, such as that caused by ultraviolet radiation. Notably, Claspin undergoes phosphorylation within its Chk1 binding domain in response to replication stress, a process that is crucial for its function. This domain features two highly conserved repeats, each approximately ten amino acids long, containing serine residues (serine 864 and serine 895) that are phosphorylated during stress conditions. The binding of Chk1 to Claspin enhances the interaction between Chk1, ATR, and Rad9, effectively halting the cell cycle to allow for DNA repair. Claspin is predominantly expressed during the S/G2 phase of the cell cycle, underscoring its importance in cellular responses to DNA damage and replication stress.
