cHMGCS Antibody (H-9) is a mouse monoclonal IgG1 antibody that detects cHMGCS in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). cHMGCS, or cytoplasmic HMG-CoA synthase, plays a crucial role in the mevalonate pathway, which is essential for the biosynthesis of isoprenoids, including cholesterol and other vital biomolecules. cHMGCS catalyzes the condensation of acetyl-CoA and acetoacetyl-CoA to form HMG-CoA, which is subsequently converted into mevalonate by HMG-CoA reductase, marking the beginning of isoprenoid biosynthesis. cHMGCS functions extend beyond cholesterol production, contributing to the synthesis of ubiquinone, dolichol, and farnesyl groups, all critical for cellular function and membrane integrity. cHMGCS is predominantly expressed in the liver and kidney, and expression is tightly regulated by nutritional states, with fasting and elevated levels of cAMP and fatty acids enhancing transcription, while feeding and insulin exert a repressive effect. The regulatory mechanisms governing cHMGCS expression involve a complex interplay of transcription factors, including PPAR, RXR, COUP-TF, and HNF-4, which bind to specific elements within the cHMGCS promoter, highlighting the enzyme′s significance in metabolic regulation and potential implications in metabolic disorders.
