CHIP Antibody (C-10) is a mouse monoclonal IgG1 kappa light chain antibody that specifically binds to the C-terminus of the human CHIP protein, recognizing amino acids 73-303. CHIP, also known as STUB1 (STIP1 homology and U-box containing protein 1), is a cytoplasmic E3 ubiquitin ligase that plays a crucial role in protein quality control by mediating the ubiquitination and subsequent degradation of misfolded or damaged proteins. Located predominantly in the cytoplasm, CHIP interacts with molecular chaperones Hsp70 and Hsp90, facilitating the degradation of their client proteins and thus maintaining cellular protein homeostasis. The cytoplasmic localization of CHIP is essential for monitoring and regulating the quality of newly synthesized proteins, preventing the accumulation of aberrant proteins that could lead to cellular dysfunction or disease. CHIP also modulates signaling pathways by interacting with proteins such as Smad1/Smad4, inhibiting BMP signaling through ubiquitin-mediated degradation of Smad proteins. Additionally, CHIP regulates the stability of oncogenic proteins like ErbB2 by controlling their association with chaperones and promoting their degradation. Expression of CHIP is most abundant in striated muscle tissues, including heart and skeletal muscle, with lower levels observed in the pancreas and brain. Anti-CHIP antibody (C-10) reacts with CHIP from mouse, rat, and human origins and is validated for use in western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA) applications.

How much sample do you recommend loading on the gel for Western blot detection using CHIP (C-10): sc-133083?
