

Chibby Antibody (G-4) is a mouse monoclonal IgG1 antibody that detects Chibby in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Chibby, also known as Cytosolic leucine-rich protein or PIGEA-14, is a highly conserved protein consisting of 126 amino acids that plays a crucial role in inhibiting β-catenin-mediated transcriptional activation by competing with LEF-1 for binding to β-catenin. This function is particularly important as it helps regulate the Wnt signaling pathway, which is vital for various cellular processes, including cell proliferation and differentiation. Chibby is primarily localized in the nucleus and the Golgi apparatus within the trans-Golgi network, where Chibby acts as a homodimer. Interaction with the 14-3-3 protein leads to the sequestration of Chibby in the cytoplasm, forming a stable complex with β-catenin that facilitates the nuclear export of β-catenin, thereby modulating β-catenin activity. While Chibby is widely expressed, Chibby is found at elevated levels in tissues such as skeletal muscle, heart, placenta, and kidney. Notably, down-regulation of Chibby has been observed in thyroid and metastatic uterine tumors, indicating Chibby′s potential role as a tumor suppressor.
