Cdc6 Antibody (180.2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cdc6 of mouse, rat, and human origin through western blotting, immunoprecipitation, immunofluorescence, and immunohistochemistry. Cdc6 (180.2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Cdc6 plays a crucial role in the initiation of DNA replication, which is essential for cell division and proper progression of the cell cycle. By ensuring DNA replication occurs at the right time, Cdc6 helps maintain genomic stability and prevents errors that could lead to cell malfunction or disease. Cdc6 is particularly important in cancer research, as dysregulation of Cdc6 can contribute to uncontrolled cell proliferation. Cdc6 is the human homolog of Saccharomyces cerevisiae Cdc6, and interaction with various proteins, such as Cdc37, aids in the formation of the cyclin D1/Cdk4 complex, highlighting an integral role in cell cycle regulation. Additionally, Cdc6 interacts with ubiquitin-conjugating enzymes like Cdc34, Cdc27, and Cdc16, which are involved in the degradation of cyclins, further emphasizing importance in the orchestration of cell cycle events.

Thanks. I'm working on paraffin-embedded mouse livers. The tissue has proven troublesome in the past (i.e. antibodies working just fine on other paraffin tissues not working at all). Any particular suggestion (i.e. harsher retrieval conditions)?

We do not have a specific protocol for mouse liver, but here is a link to our suggested protocol: https://www.scbt.com/resources/protocols/immunoperoxidase-staining We recommend sodium citrate with heat as the primary method, but you can also try any of the other methods in the protocol.

Is the antibody suitable for immunofluorescence from paraffin-embedded tissues? If so, what antigen retrieval protocol do you recommend?  Thanks in advance.

Thank you for your question. Yes, this antibody, sc-9964, Cdc6 (180.2) is recommended for IF/IHC on paraffin embedded tissue sections. We recommend a starting dilution of 1:50, and suggest antigen retrieval using sodium citrate with heat. The antibody sc9964 recognize which part of Cdc6, what is the meaning of 180.2, thank you !

Thank you for your question. The exact epitope recognized by this monoclonal antibody has not been determined. And 180.2 is the clone designation for this antibody.

How can Cdc6 Antibody (180.2): sc-9964 be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This mouse monoclonal antibody is currently available phycoerythrin (sc-9964 PE), fluorescein (sc-9964 FITC), Alexa Fluor® 488 (sc-9964 AF488) or Alexa Fluor® 647 (sc-9964 AF647).
