CD45RO Antibody (UCH-L1) is a mouse monoclonal IgG2a antibody engineered to target the CD45RO isoform of the CD45 protein, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC). CD45RO is a crucial transmembrane glycoprotein expressed predominantly on memory T lymphocytes in humans, as well as on various hematopoietic cells in mice and rats, playing a vital role in regulating immune cell signaling and activation. Structurally, CD45RO consists of an extracellular domain generated through alternative splicing of exons 4, 5, and 6, resulting in a shorter extracellular region compared to other isoforms like CD45RA. This structural variability allows CD45RO to interact differently with other cell surface molecules, thereby modulating essential signal transduction pathways involved in T cell receptor (TCR) signaling. Located on the cell membrane, CD45RO functions as a phosphotyrosine phosphatase, dephosphorylating key signaling proteins such as Src family kinases Lck and Fyn, as well as Syk family kinases like ZAP-70. These interactions are pivotal for initiating and sustaining signaling cascades that drive T cell activation, proliferation, and differentiation, which are critical for maintaining immune system homeostasis and effective immune responses. Additionally, CD45RO undergoes post-translational modifications, including extensive glycosylation, which are essential for its stability, proper membrane localization, and interactions with other signaling proteins, thereby enhancing its regulatory functions. Anti-CD45RO antibody (UCH-L1) is raised against cultured human T cell line CA-1 and is effective in various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM) across human, mouse, and rat samples. By binding to CD45RO, CD45RO monoclonal antibody (UCH-L1) facilitates comprehensive analysis of memory T cell populations and their interactions with key signaling proteins, providing valuable insights into immune system dynamics and the mechanisms underlying immune-related diseases.
