CD45 Antibody (35-Z6) is a mouse monoclonal IgG1 kappa light chain antibody designed to detect CD45, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC), in mouse, rat, and human samples through western blotting (WB), immunoprecipitation (IP), PAR assays, and flow cytometry (FCM). Anti-CD45 antibody (35-Z6) targets the extracellular domain of human CD45, ensuring accurate recognition across multiple species. CD45 is a crucial transmembrane glycoprotein broadly expressed on hematopoietic cells, where it plays a vital role in regulating immune cell signaling. By functioning as a phosphotyrosine phosphatase, CD45 modulates the activity of Src and Syk family kinases, such as Lck, Fyn, and ZAP-70, which are essential for initiating signaling cascades following T and B cell receptor engagement. This regulation is fundamental for effective T cell activation, proliferation, and differentiation, as well as for B cell development and antibody production. The diverse isoforms of CD45, resulting from alternative splicing of exons 4, 5, and 6, allow for fine-tuned control of immune responses tailored to specific cell types and stages of activation. Dysregulation of CD45 has been implicated in various immune disorders, including autoimmune diseases and leukemias, highlighting its importance in maintaining immune homeostasis and its potential as a therapeutic target. CD45 (35-Z6) antibody is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and Alexa Fluor® conjugates, providing versatile options for a wide range of experimental applications. Anti-CD45 antibody (35-Z6) can effectively study the expression, isoform distribution, and functional dynamics of CD45, thereby advancing our understanding of immune cell biology and related disease mechanisms.

Can this antibody detect CD45 in Zebrafish?

CD45 has only been validated for Mouse, Rat, and Human. CD45 has no similar homology to zebrafish.

Does this antibody detect WBC from C57Bl/6 mice?

If I am going to stain mouse skin tissues, do I need additional kit, for example "mouse on mouse kit" (as this antibody is mouse origin)?

Thank you for your question. It depends on what type of tissues you are staining. If you are staining mouse skin tissues, and there is not a lot of endogenous mouse IgG, then it is probably not necessary to use a special "mouse on mouse kit" for staining. We do recommend including a negative control tissue where you omit primary antibody and stain with secondary antibody alone. Then you can identify any possible non-specific binding from your anti-mouse IgG secondary antibody alone. As an alternative, you could also consider using one of the directly conjugated forms available of this CD45 antibody. Then no secondary antibody is needed for detection, and you can avoid any possible problem of non-specific binding from anti-mouse IgG secondary antibodies.

What could be the expiry date for CD45 since we have observed it to be active even after 5 years

Thank you for your question. We do not give our antibodies an expiration date since they can work for up to 10-15 years in some cases. We warranty the performance of our antibody reagents for 1 year from the date of purchase.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with CD45 Antibody (35-Z6): sc-1178?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
