SEE ALSO...

CD45RB Antibody (PD7/26) is a mouse monoclonal IgG1 kappa light chain antibody designed to target the CD45RB isoform of CD45, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC). CD45RB is a crucial transmembrane glycoprotein expressed on the surface of all nucleated hematopoietic cells in humans, playing an essential role in regulating immune cell signaling and activation. Structurally, CD45RB comprises an extracellular domain generated through alternative splicing of exons 4, 5, and 6, a single transmembrane region, and a conserved intracellular domain that possesses phosphatase activity. This structural organization enables CD45RB to interact differently with other cell surface molecules, thereby modulating key signal transduction pathways involved in T cell receptor (TCR) and CD3 complex signaling. Located on the cell membrane, CD45RB dephosphorylates specific tyrosine residues on Src family kinases such as Lck and Fyn, as well as on Syk family kinases like ZAP-70, which are essential for initiating and sustaining signaling cascades that drive T cell activation, proliferation, and differentiation. Additionally, CD45RB undergoes significant post-translational modifications, including extensive glycosylation, which are crucial for stability, proper localization on the cell membrane, and interactions with other signaling proteins. CD45RB monoclonal antibody (PD7/26) is raised against human peripheral blood lymphocytes (PBL) maintained in T-cell growth factor and is effective in various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM) across human, mouse, and rat samples. CD45RB (PD7/26) antibody facilitates comprehensive analysis of immune cell populations and their interactions with key signaling proteins, providing valuable insights into immune system dynamics and mechanisms underlying immune-related diseases. Anti-CD45RB antibody (PD7/26) can detect multiple isoforms of CD45RB, enabling researchers to investigate the diversity and function of different immune cell subsets, making this tool indispensable for both research and diagnostic purposes in immunology and hematology.

How can CD45RB (PD7/26): sc-53203 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: Randy McDonald
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-53203 PE) or fluoroscein (sc-53203 FITC).
