CD137 Antibody (BBK-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD137 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). CD137 (BBK-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD137 protein, also known as ILA and 4-1BB in mouse, is a member of the tumor necrosis factor receptor family and plays a crucial role in the immune response by delivering a costimulatory signal to T lymphocytes. CD137 is primarily expressed on activated T cells and interacts with an inducible ligand found on B cells, macrophages, and dendritic cells, which is essential for effective antigen presentation and the generation of cytotoxic T cells. Notably, crosslinking of CD137 ligand can induce apoptosis in resting lymphocytes, while CD137 promotes survival of peripheral monocytes by inducing a specific cytokine release profile, mediated by macrophage colony-stimulating factor and, to a lesser extent, granulocyte-macrophage colony-stimulating factor and interleukin-3. Additionally, soluble forms of CD137 have been detected in sera of patients with rheumatoid arthritis, suggesting a potential role in modulating immune responses and providing a negative feedback mechanism.

How much protein do you recommend loading on the gel for Western blot detection using CD137 (BBK-2): sc-58947 monoclonal antibody?

Rated 5 out of 5 by from WBのバンドがクリア免疫沈降したサンプルのウエスタンブロットで、バンドがとてもクリアに出ていました。熱処理してあるはずのsampleでしたが、dimerの位置にバンドを薄く認めました。バンドの位置は、32kDa よりは少し下側に、あと少し気になったのは、50kDa辺りにも、バンドがあることでしょうか。
