    cathepsin W Antibody (1B1) is a mouse monoclonal IgG1 kappa light chain antibody that detects cathepsin W of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-cathepsin W antibody (1B1) is available as a non-conjugated format. Cathepsin W, also known as lymphopain, is part of a unique subgroup of cathepsin proteases, which includes cathepsin F, and is phylogenetically distinct from other human cathepsins. The cathepsin W gene is located on chromosome 11q13.1 and consists of ten exons with introns that vary in size from 81 to 119 base pairs. This protein is specifically expressed in CD8+ T lymphocytes, marking its critical role in the immune response. The expression of cathepsin W begins during the differentiation of thyrocytes into CD8+ T lymphocytes, coinciding with the downregulation of CD4+ receptor expression in thymocytes. Notably, cathepsin W is localized within the rough endoplasmic reticulum in transfected Cos-7 and HeLa cells, which is essential for proper folding and function. Cathepsin W features a unique 21 amino acid peptide insertion between the active site histidine and asparagine residues, along with a distinctive 8-amino acid carboxy-terminal extension. Additionally, cathepsin W exhibits an extended loop structure in the second or beta-sheet domain and possesses an extra disulfide bond, which are signature characteristics that contribute to stability and activity. Furthermore, cathepsin W may exist as a dimer, with each monomer linked by a disulfide bond, highlighting complex structural dynamics that are crucial for enzymatic function in immune regulation.
