cathepsin D Antibody (D-7) is a mouse monoclonal IgM kappa antibody targeting an epitope mapped between amino acids 383-411 at the C-terminus of human cathepsin D, with reactivity in mouse, rat, and human species. Anti-cathepsin D antibody (D-7) is validated for applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Cathepsin D, also known as CTSD, is a lysosomal aspartyl protease involved in intracellular protein degradation and processing protein precursors into bioactive peptides. Cathepsin D plays a critical role in cellular metabolism, autophagy, and apoptosis, making cathepsin D essential for maintaining cellular homeostasis. As a zymogen, cathepsin D undergoes proteolytic cleavage to become active, and this process is tightly regulated to ensure proper function within the acidic lysosomal environment. Cathepsin D activity influences key biological processes, including tumor progression, where cathepsin D can contribute to cancer cell invasion and metastasis, as well as neurodegenerative diseases. Cathepsin D interacts with proteins involved in stress response and apoptotic pathways, further emphasizing cathepsin D′s regulatory role. Expression of cathepsin D is regulated by transcription factors such as Sp1 and AP-2, which bind to cathepsin D′s promoter region in response to cellular demands. Anti-cathepsin D antibody (D-7) is an invaluable tool for exploring the functional importance of cathepsin D in health and disease.

Thank you for your question. We validate this antibody for mouse, rat and human. There is a 78% homology in pig. We only honor our warranty if the homology is 95% or higher.

Does this work by immunofluorescence in mouse cells? Is there a recommended dilution for this application? Thank you.

Anti-cathepsin D Antibody (D-7): sc-377299 is recommended for detection of cathepsin D of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. For immunofluorescence, the recommended starting dilution is 1:50, with a dilution range between 1:50-1:500.

What is the best reagent to dilute this antibody for Western Blotting?

Thank you for your question. We recommend using our TBS Blotto A: sc-2333 (https://www.scbt.com/scbt/product/tbs-blotto-a) which is 5% nonfat milk + 1X TBS + 0.05% Tween-20. This is the recommended buffer for blocking, as well as primary and secondary antibody incubations for Western blotting.

cathepsin D (D-7): sc-377299 is a mouse IgM, can I use an anti-mouse IgG secondary antibody for detection?

We recommend using an isotype-specific anti-mouse IgM secondary antibody for detection of this mouse IgM primary antibody. Anti-mouse IgG secondary antibodies may work, due to the similar structures of IgG and IgM, but they are not ideally suited.

Rated 4 out of 5 by from It works wellWe checked this antibody on lysates from immortalized human proximal tubular epithelial cells by Western blot. It detects a single band (around 48-50 kDa, presumably the immature form) with no background signals, after overnight incubation (1:500) at 4°C. We were unable to detect the mature form of Cathepsin D (35 kDa) in these cells upon these experimental conditions.

Rated 5 out of 5 by from works very well for mouse fat tissueI did 1/500 dilution for 4 degree overnight incubation, which gives me expected band, upper band between 40-55 kDa, around 52Kda or 46 kDa, I am not sure. But it also can detect the mouse IgG light chain, the bottom band
