Calregulin Antibody (A-9) is a mouse monoclonal IgG1 kappa light chain antibody that detects Calregulin in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Calregulin antibody (A-9) is offered in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Calregulin, also known as calreticulin, is a calcium-binding protein predominantly located in the lumen of the endoplasmic reticulum, where Calregulin plays a crucial role in protein folding and quality control. Calregulin′s ability to bind calcium is vital for function, as Calregulin helps regulate calcium homeostasis within the cell, which is essential for various cellular processes, including signal transduction and muscle contraction. The presence of both low- and high-affinity calcium-binding sites allows Calregulin to effectively modulate calcium levels, thereby influencing the activity of other proteins and cellular pathways. Calregulin interacts with several key proteins involved in glycoprotein maturation, ensuring that only properly folded proteins are transported to their final destinations, which is critical for maintaining cellular health and function.

I want to detect calreticulin on the cell surface of mouse cells by a conjugated antibody. There are two choices for this aim, conjugated Calregulin (A-9) and (F-4). which one of them, (A-9) or (F-4), is more suitable for cell surface calreticulin?

Thank you for your question. Both antibodies are recommended for the detection of murine calregulin by IF or IHC(P). For Calregulin Antibody (F-4): sc-373863 there are two publications showing cell surface staining: PMID 27789271 and PMID 29324305. I am using Calregulin (A-9): sc-166837 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Rated 5 out of 5 by from Wonderful antibodyI recently tested 2 Calreticulin/Calregulin Sc-373863 and Sc-166837 antibodies in human cell lines and both of them turned equally good. Calregulin gives approximately 55kDa band and expressed widely in human tissues. HEP-G2 is a liver cancer cell line and HEK293 was derived form embryonic kidney carcinoma but the later reported to express neuronal proteins and may be neuronal in origin. Highly recomend this antibody, See my blots.

Strong, clear signal with little to no background. Beautiful tissue staining. Works well in a variety of species including mouse, rat, and human samples.
