Calnexin Antibody (TO-5) is a mouse monoclonal IgG1 kappa light chain antibody raised against a synthetic peptide corresponding to amino acids 529-544 of human Calnexin. Calnexin, also known as CANX or IP90, is a calcium-binding chaperone protein localized in the membrane of the endoplasmic reticulum (ER). Anti-Calnexin antibody (TO-5) plays a crucial role in the ER′s quality control system by interacting with newly synthesized N-linked glycoproteins, ensuring their proper folding and assembly. Calnexin binds to monoglucosylated glycoproteins via its lectin domain, retaining them in the ER until they achieve correct conformation. Post-translational modifications of Calnexin, such as phosphorylation, are important for regulating its chaperone activity and interactions with substrate proteins. Phosphorylation can modulate its affinity for substrates and influence ER stress responses and protein homeostasis. Proper functioning of Calnexin is essential for preventing the accumulation of misfolded proteins, which can lead to diseases like neurodegeneration and diabetes. Calnexin monoclonal antibody (TO-5) reacts with human Calnexin and is suitable for applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM), providing a valuable tool for researchers studying protein folding and ER quality control mechanisms.

Do you have any data demonstrating the cross-reactivity of the anti-Calnexin clone TO-5 to mouse cells or tissue? The paper you reference for WB on a mouse cell line (pubmed ID 23660595) shows no calnexin bands ... the data all negative.

Thank you for your question. We do not currently have any data using this monoclonal antibody, Calnexin (TO-5): sc-80645, in mouse or rat samples. We will reevaluate the species reactivity for this product and update our product information accordingly. If you have already purchased the antibody, please contact our Technical Service Department and we can support your purchase for any current recommended species per our product warranty. We will remove the product citation you mentioned for this antibody, as in the paper it does indicate that “Immunoreactive calnexin bands were not seen in any of the plasma membranes examined.” As an alternative, we would recommend using our alternate antibody, Calnexin (E-10): sc-46669. Can Calnexin (TO-5): sc-80645 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Calnexin (TO-5): sc-80645 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
